Olaparib is a new oral polyadenosine diphosphate ribose polymerase [PARP] inhibitor. PARP includes the most important members of three protein families: PChemicalbookARP1, PARP2 and PARP3. PARP enzymes are involved in normal cellular homeostasis such as DNA transcription, cell cycle regulation and DNA repair.

Olaparib is a potent poly(ADP-ribose) polymerase (PARP) inhibitor. Olaparib has been shown to induce significant killing of ATM-deficient lymphoid bad cells in vitro and in vivo. Recent studies show that Olaparib increases radiosensitivity of a lung disease xenograft, making it a potential candidate for use in combination with radiotherapy.
