Larotinib is an inhibitor of tropomyosin receptor kinase (Trk), TrkA, TrkB and trkC. TrkA, B and C are encoded by genes ntrk1, NTRK2 and ntrk3. They can produce a chimeric Trk fusion protein that can be activated. The protein can be used as a carcinogenic driver to promote cell proliferation and survival in tumor cell lines. Larotinib shows antitumor activity in cells with constitutive activation of Trk protein, deletion of protein regulatory domain or overexpression of Trk protein caused by gene fusion
