Loxo-101 is also known as larotinib. Larotrectinib (LOXO-101) was approved by the U.S. Food and Drug Administration (FDA) in 2018 to treat adults and children with locally advanced or metastatic solid tumors with NTRK gene fusion. Loxo-101, an antitumor drug targeting tropomyosin kinase (TRK), was approved by the US FOOD and Drug Administration (FDA) on November 26, 2018. The efficacy of LOXO-101 was evaluated based on three multicentre, open-label, single-group clinical trials in which treatment was evaluated by measuring the total response rate and response time. In 55 patients with unresectable or metastatic solid tumors with NTRK gene fusion identified by high-throughput sequencing (NGS) or fluorescence in situ hybridization (FISH), loXO-101100mg was orally administered twice daily in adults and 100mg•m-2 on the body surface in pediatric patients (18 years of age or younger), with a maximum dose of 100mg. Twice a day. Clinical data showed that loXO-101 had a total response rate of 75% in patients with different types of solid tumors with NTRK gene fusion, while Chemicalbook had a complete response rate of 22% and a partial response rate of 53%. 73% of the patients had a response time of more than 6 months, and 39% had a response time lasting more than a year. In the latest clinical data presented at the 2018 European Medical Oncology Conference, LoxoOncology presented the latest clinical data for the treatment of TRK fusion cancer patients with LOXO-101. In the 67 newly reported TRK fusion patients, the total response rate was 81%, of which the partial response rate was 63%, and the complete response rate was 17%, showing good antitumor activity. The most common adverse reactions (≥20%) to LOXO-101 were fatigue, nausea, dizziness, vomiting, elevated AST, cough, elevated ALT, constipation, and diarrhea.
